In the comments to the article "Exchange 2016: Install cumulative updates (CU)" I was informed that with Veeam Backup & Replication 9.5 U1 and Exchange 2016 CU5 there are problems with the single item restore. The databases can be backed up and restored normally, but the Veeam Explorer for Exchange no longer works with Exchange 2016 CU5 databases.
Mailboxes can no longer be displayed in Veeam Explorer, so it is not possible to restore individual mails or mailboxes. Expanding the database does not work:

Updating the ESE.dll did not help me. Apparently the problem is caused by changes to the Exchange 2016 CU5 database structure.
Since CU5, other backup tools can also no longer perform a single item restore, here for example Veritas:

As a workaround, however, the database can still be restored to a recovery database. Individual mailboxes can then be restored or exported from the recovery database using Exchange on-board tools.
Veeam will fix the problem with the U2, which is already in the starting blocks. Many thanks to Ralle for the tip!
